ABSTRACT

OBJECTIVE@#To explore the similarities and variations of biological phenotype and cytotoxicity of human umbilical cord blood natural killer cells (hUC- NK) after human umbilical cord blood-derived mononuclear cells (hUC-MNC) activated and expanded by two in vitro high-efficient strategies.@*METHODS@#Umbilical cord blood mononuclear cells (MNC) from healthy donor were enriched by Ficoll-based density gradient centrifugation. Then, the phenotype, subpopulations, cell viability and cytotoxicity of NK cells derived from Miltenyi medium (denoted as M-NK) and X-VIVO 15 (denoted as X-NK) were compared using a "3IL" strategy.@*RESULTS@#After a 14-day's culture, the contents of CD3-CD56+ NK cells were elevated from 4.25%±0.04% (d 0) to 71%±0.18% (M-NK) and 75.2%±1.1% (X-NK) respectively. Compared with X-NK group, the proportion of CD3+CD4+ T cells and CD3+CD56+ NKT cells in M-NK group decreased significantly. The percentages of CD16+, NKG2D+, NKp44+, CD25+ NK cells in X-NK group was higher than those in the M-NK group, while the total number of expanded NK cells in X-NK group was half of that in M-NK group. There were no significant differences between X-NK and M-NK groups in cell proliferation and cell cycle, except for the lower percentage of Annexin V+ apoptotic cells in M-NK group. Compared with X-NK group, the proportion of CD107a+ NK cells in M-NK group were higher under the same effector-target ratio (E∶T) (P<0.05).@*CONCLUSION@#The two strategies were adequate for high-efficient generation of NK cells with high level of activation in vitro, however, there are differences in biological phenotypes and tumor cytotoxicity.

ABSTRACT

Objective: To investigate the effects of continuous exercise training (CT) and high-intensity interval exercise training (HIIT) on liver lipid metabolism and the correlation of the level of fibroblast growth factor 21(FGF21) in serum and liver tissues. Methods: Male SD rats were randomly divided into normal diet group (N) and obesity model group (H) after 1 week of adaptive feeding. Rats in the obesity model group were fed with 45% high-fat diet for about 8 weeks, and 20% weight increase compared with normal rats was considered as obesity. The rats were divided into normal diet control group (LC), normal diet HIIT group (LHI), normal diet CT group (LCT), High fat diet-induced obese control group (OC), obese HIIT group (OHI), and obese CT group (OCT) (n=10). Exercised rats were given weight-bearing swimming training intervention for 8 weeks. Blood samples were collected at least 24h after the last exercise intervention to detect the serum levels of inflammatory factors and FGF21. Liver tissue samples were collected to detect the lipid content, lipid metabolic enzyme content and FGF21 expression level. Results: Compared with LC group, the body weight, serum inflammatory factors levels and hepatic triglyceride content were increased significantly (P<0.05). Hepatic triglyceride content was downregulated in LHI group and FGF21 expression level was enhanced in LCT group (P<0.05). Compared with OC group, the body weight and hepatic triglyceride content were decreased significantly (P<0.05), mitochondrial CPT-1β and β-HAD enzyme contents in liver were increased significantly (P<0.05) in OHI group, the contents of LPL and FAT/CD36 enzyme in liver and the levels of FGF21 in serum and liver of OCT group were increased significantly (P<0.05). Conclusion: Both exercise modes can reduce the body weight in normal and obese rats, and lipid deposition in the liver of obese rats. HIIT has a more significant effect on alleviating liver lipid deposition in obese rats by upregulating mitochondrial lipid oxidation level in normal and obese rats. CT improves the levels of FGF21 in serum and liver tissues of normal and obese rats, enhances enzyme contents that involved in fatty acids uptake to the liver, which has limited effect on alleviating lipid deposition in liver of obese rats.

ABSTRACT

OBJECTIVE:To study the intervention eff ects and pot ential m echanism of celastrol on non-alcoholic steatohepatitis (NASH)induced by methionine-choline deficiency (MCD)diet. METHODS :Male C 57BL/6J mice were randomly divided into normal control group ,model group ,celastrol low-dose and high-dose groups [ 0.5,1 mg/(kg·d)],with 7 mice in each group. The normal control group was given a methionine-choline sufficient diet ,while the model group and administration groups were fed an MCD diet to induce NASH model. At the same time ,normal control group and model group were given polyoxyethylene castor oil intragastrically;administration groups were given relevant drugs intragastrically ;the volume of gavage was 0.1 mL/g,once a day , for consecutive 4 weeks. The liver morphology was observed ,and the pathological changes of liver tissue were observed by HE staining and oil red O staining. The levels of serum liver enzymes (AST,ALT),and the levels of lipid indexes (TC,TG)in serum and liver tissue were detected by enzyme method. The protein expression of NF-κB p65,TNF-α and IL-6 in liver tissue were determined by Western blotting assay. RESULTS :Compared with normal control group ,the volume of the liver was reduced and the color was yellow ,and the surface was rough in model group ;inflammatory cell infiltration ,fat vacuoles and lipid droplets aggregation were found in the liver tissue ;the serum levels of TC and TG were significantly decreased ,the levels of serum liver enzymes and protein expression of NF-κB p65,TNF-α and IL-6 in liver tissue were significantly increased (P<0.01). Compared with model group ,the liver surface of each administration group was ruddy and smooth without brown spots ,the inflammatory cells and fat vacuoles in liver tissue were reduced ,and the coverage area of lipid droplets was reduced ;the levels of serum TC and TG were significantly increased ,the levels of serum liver enzymes ,the levels of TG and protein expression of NF-κB,TNF-α and IL-6(except for celastrol low-dose group )in liver tissue were significantly decreased (P<0.05 or P<0.01). CONCLUSIONS : Celastrol can improve the liver injury of NASH model mice induced by MCD diet ,which is related to the reduction of TG accumulation in liver tissue and inhibition of the expression of inflammatory related factors.

ABSTRACT

Objective:To investigate the correlation between serum procalcitonin (PCT) level and intracranial atherosclerotic burden (ICASB) in patients with ischemic stroke. Methods:From January 2019 to December 2020, consecutive patients with acute ischemic stroke admitted to the Department of Neurology, Jiangsu Provincial Second Chinese Medicine Hospital were enrolled. Chemiluminescence immunoassay was used to detect serum PCT levels, and ICASB was evaluated based on the results of cranial magnetic resonance angiography. Univariate analysis was used to determine the baseline data among the different ICASB score groups. Then the independent correlation between serum PCT level and ICASB was determined by the ordinal logistic regression analysis. At the same time, the correlation between serum PCT level and ICASB was determined by the linear regression analysis. Results:A total of 291 patients with acute ischemic stroke were enrolled, including 161 male (55.3%), aged 64.5±8.4 years; median serum PCT level was 0.053 μg/L. According to the ICASB score, the patients were divided into 0 group ( n=155, 53.3%), 1-3 group ( n=95, 32.6%) and >3 group ( n=41, 14.1%). Univariate analysis showed that the age, serum homocysteine and PCT level, as well as the proportion of diabetes were significantly higher in the higher ICASB score group, while the proportion of the patients with atrial fibrillation was significantly lower (all P<0.05). Ordinal multivariable logistic regression analysis showed that higher serum PCT level was an independent factor for higher ICASB score (the 4 th quartile vs. the 1 st quartile: odds ratio, 2.015, 95% confidence interval 1.052-3.927; P=0.043). Multiple linear regression analysis showed that the serum PCT level was positively correlated with the ICASB score ( r=0.253, P=0.001). Conclusion:The serum PCT level is correlated with ICASB.

ABSTRACT

OBJECTIVE@#To explore the effect of miR-335-5p/ADCY3 interaction on the lymphocyte function in the patients with aplastic anemia (AA).@*METHODS@#Blood samples were collected from 22 healthy volunteers (HC) and 50 AA patients including 38 severe AA (SAA) and 12 non-severe AA (NSAA). Peripheral blood mononuclear cells (PBMNC) were isolated. The expression of miR-335-5p and ADCY3 mRNA was detected by using RT-PCR. Negative control miR-335-5p (NC group) and miR-335-5p mimic (mimic group) were transfected to AA-PBMNC by using RNAimax reagent, respectively. The proliferative ability, activation and cytokines of CD4 T and CD8 T cells were measured by flow cytometry. Dual-luciferase reporter assay was used to verify the targeted relationship between miR-335-5p and target gene.@*RESULTS@#The expression of miR-335-5p was significantly downregulated in SAA-PBMNC and NSAA-PBMNC compared with HC-PBMNC (0.08±0.01 vs 0.74±0.10, P<0.01; 0.17±0.02 vs 0.74±0.10, P<0.01). Meanwhile, the expression of miR-335-5p in SAA-PBMNC was very statistically significantly lower than that in NSAA-PBMNC (P<0.01). Compared with NC group, upregulation of miR-335-5p in vitro could significantly inhibited the proliferation of CD4 T and CD8 T cells in AA-PBMNC (P<0.05 and P<0.05, respectively). And, upregulating miR-335-5p in AA-PBMNC could significantly inhibited the activation of CD4 and CD8 T cells (P<0.01 and P<0.01, respectively). The ratio of CD4TNFα T, CD8IFNγ+T and CD8TNFα T cell by up-regulating the expression of miR-335-5p from AA-PBMNC in vitro was also significantly lower (P<0.01, P<0.05 and P<0.05, respectively). In addition, the expression of ADCY3 was higher in AA-PBMNC than that in HC-PBMNC (1.70±0.15 vs 0.76±0.12, P<0.01). Furthermore, by means of dual-luciferase reporter assay, the luciferase activity of ADCY3'UTR wildtype could be inhibited by miR-335-5p.@*CONCLUSIONS@#The expression of miR-335-5p was significantly downregulated in AA, and that correlates with disease severity. Up-regulating miR-335-5p can correct the hyperimmune status in AA patients by targeting ADCY3. These changes may relates with the strengthen of inhibition for targeted gene ADCY3.

ABSTRACT

OBJECTIVE: To study the improvement effect and mechanism of methylated urolithin A on oleic acid-induced lipid accumulation in human liver cancer Huh-7 cells. METHODS: Oleic acid was adopted to induce lipid accumulation model cells. Huh-7 cells were divided into control group (culture medium), model group (1 mmol/L oleic acid), low-dose group (1 mmol/L oleic acid+10 μmol/L methylated urolithin A) and high-dose group (1 mmol/L oleic acid+20 μmol/L methylated urolithin A). Oil red O staining was used to observe lipid accumulation in cells. Triglyceride(TG) enzyme assay was applied to determine the TG content in cells. PCR was employed to detect the mRNA expression of FASN, SREBP-1, PPAR-α and PPAR-γ in cells. Western blotting was used to determine the protein expression of FASN in cells. RESULTS: After induced by oleic acid, a large amount of lipid droplet accumulated around the cells; the intracellular lipid and TG content, mRNA expression levels of FASN, SREBP-1 and PPAR-γ, protein expression levels of FASN were increased significantly, while mRNA expression level of PPAR-α was decreased significantly (P<0.01). After intervened with methylated urolithin A, lipid droplet around the cells decreased significantly; the contents of lipid and TG in cells were decreased significantly, while the mRNA expression levels of FASN, SREBP-1 and PPARγ and protein expression level of FASN were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: Methylated urolithin A can improve oleic acid-induced lipid accumulation in Huh-7 cells, the mechanism of which may be associated with inhibiting fat synthesis, promoting lipid metabolism and down-regulating the expression of metabolism-related factors as FASN, SREBP-1 and PPAR-γ.

ABSTRACT

Objective The abnormal ankle-brachial index ( ABI) is associated with the incidence of cardiocerebral vascular diseases, but little is known about its relationship with cerebral microbleeds (CMB).This study aimed to investigate the correlation be-tween ABI≤0.9 and different distribution patterns of CMB . Methods We enrolled 187 patients with acute lacunar infarction , inclu-ding 115 non-CMB cases and 72 CMB cases (20 strictly lobar, 24 strictly deep, and 28 lobar and deep). We analyzed the differences between the two groups and the association of abnormal ABI with the occurrence and distribution of CMB by logistic regression analysis . Results ABI≤0.9 was found in 57 (30.5%) of the patients, with a significantly higher incidence rate in the CMB group than in the non-CMB group (43.1%vs 22.6%, P=0.003).The level of ABI was negatively correlated with the number of CMBs (r=-0.211, P=0.006).Multivariate logistic regression analysis after adjusted for confounders indicated that ABI ≤0.9 was significantly associated with the presence of CMB (OR=2.363;95%CI:1.181-4.729), deep CMB (OR=3.434;95%CI:1.283-9.187), and lobar and deep CMB ( OR=2.837;95%CI:1.098-7.333) in patients with ischemic cerebrovascular disease . Conclusion Decreased ABI is a risk factor of CMB, particularly deep CMB, in patients with ischemic stroke.

Objective To assess the immune level of cervical cancer patients and precancerous lesions patients, and investigate the association of Th 1 /Th2 type cytokines expression and clinical pathologic factors in the plasma of Uyghur cervical cancer patients .Methods we collected peripheral blood specimens from cervical cancer patients, precancerous lesion(CINⅢ)patients and controls. The expressions of Th1 type cytokines IL-2, IFN-γand Th2 type cytokines IL-4,IL-10 in plasma were detected by ELISA .Receiver operating characteris-tic curve( ROC curve) was used to analyze the cytokines value of auxiliary diagnosis in cervical cancer .Results The results showed that compared with control group ,the expressions of Th1 type cytokines IL-2,IFN-γin cer-vical cancer and precancerous lesion group were significantly reduced (P<0.05).The expressions of Th2 type cytokines IL -4, IL -10 in cervical cancer and precancerous lesion group were significantly increased ( P <0.05).in cervical cancer group,IL -10 expression gradually increased with tumor pathological staging (P <0.05).IL-2 expression level in Uyghur cervical cancer patients was significantly lower than in Han patients (P<0.05).Compared cervical cancer group with control group ,AUC of IL-2,IFN-γ,IL-4 and IL-10 were 0.979,0.766,0.736 and 0.903.Conclusion Cellular immune level of cervical cancer patients was low and Th1/Th2 shift has occurred , which suggests that it may be one of the mechanisms in immune escape of tumor cells.Th1 /Th2 type cytokines detection has a certain significance for auxiliary diagnosis of cervical cancer .Be-sides,the decrease of IL-2 expression may play an important role in the occurrence and development of Uyghur patients with cervical cancer .

ABSTRACT

Objective Depression is a mood disorder that causes a persistent feeling of sadness, with high morbidity rates and great social impairment.Increasingly studies show the abnormalities of brain networks. We summarized the results of resting state functional magnetic resonance imaging study of depression, and demonstrated the neural loops mechanism from neuroimaing perspective. Methods The key words depression, resting state and network were searched in PubMed,CNKI and Wan Fang databases from January 2000 to December 2014.The nodes of depression related network and the alterations of cortex resting-state networks were summarized. Results 24 studies focusing on resting state network of depression were identified.40 studies based on ROI (region of interest) analysis, which included amygdala, frontal lobe, pregenual anterior cingulate cortex and cerebellum. The functional connectivity of ROIs were calculated and compared between groups.8 studies based on ICA (independent component analysis),the resting state networks were extracted and compared between groups. Two based on graph theory, the functional connectivity of whole brain were analyzed and compared. Conclusion There are abnormalities of functional connectivity among limbic system-thalamus-frontal cortex, and the changes of functional connectivity were associated with clinical symptom and drug efficacy of depression.

ABSTRACT

Objective]With discussion of stroke machine with depression syndrome and law,we broaden the clinical thinking and improve clinical therapeutic effect.[Methods]Ancient medical books discussion of stroke and depression syndrome were collected, referring to the definition of post-stroke depression in modern medicine, traditional Chinese medicine continued to collect relevant papers in modern treatises, and combined with the experience of the author's departments in the diagnosis and treatment of post-stroke depression.[Outcome]Post-stroke depression in the pathogenesis of disorders with blood and viscera of yin and yang, phlegm and qi stagnation as the standard.Treatment should be given on multi-organs,especially focusing on regulating liver;emphasis interconversion between pathological factors in clinic, adjusting for the first Qi; taking care treatments with acupuncture combined emotions inter-resistance. Through organizing the relevant literature depression after stroke, come to the disease and its treatment of its laws,as well as the clinical treatment of attention points, which can improve clinical outcomes.[Conclusion] The pathogenesis of post-stroke depression is varied, we must analyse the syndrome and law carefully to promote better recovery, and then improve clinical therapeutic effect.

ABSTRACT

INTRODUCTIONProximal femoral nail antirotation (PFNA) and third-generation Gamma nail (Gamma 3) are widely used in the treatment of intertrochanteric fractures. However, it remains unclear which device achieves better clinical and radiographic outcomes when treating intertrochanteric fractures.

METHODSThis study comprised 239 patients with intertrochanteric fractures treated with either PFNA or Gamma 3 for a minimum of 12 months. During surgery, the operative time, image intensifier time and amount of blood loss were recorded. Following surgery, we assessed reduction quality and implant position. At the final follow-up, postoperative complications, including femoral shaft fracture, cutout, reoperation, pneumonia, urinary tract infection, cerebral infarction, cardiac infarction and decubital ulcer, were recorded. In addition, walking ability was assessed using the Parker-Palmer mobility score.

RESULTSNo difference was found in the operative time, image intensifier time and amount of blood loss between patients treated with PFNA and those treated with Gamma 3. The reduction quality of fractures treated with Gamma 3 was better than those treated with PFNA. However, there were no significant differences in implant position, walking ability and postoperative complications between the two groups. Although Gamma 3 resulted in better reduction quality, it did not provide any advantages in walking ability and postoperative complications when compared with PFNA.

CONCLUSIONTherefore, we conclude that both PFNA and Gamma 3 are safe and reliable devices for the treatment of intertrochanteric fractures.
